Description: The cytosolic NFκB/Rel transcription factors are in inactive state, complexed with the inhibitory IκB proteins. Phosphorylation-induced, proteasome-mediated degradation of IκB triggers their activation. The key regulatory step in this pathway involves activation of IκB kinase (IKK) complex, whose catalysis is generally carried out by three tightly associated IKK subunits. IKKγ and IKKβ serve as the catalytic subunits of the kinase. IKK serves as the regulatory subunit. Phosphorylation of serines 177 and 181 in the activation loop of IKKβ (176 and 180 in IKKα) causes conformational changes resulting in activation of IKK. Anti- IKKγ antibody can be used for IKK2/β kinase assay from immunoprecipitated (IP) cell extracts. This antibody is produced in rabbit by repeated injections of Immunogen.
