PNK (polynucleotide kinase) is a DNA repair enzyme that participates in single strand break repair and non-homologous end rejoining (NHEJ) for double strand breaks. PNK possesses a 5’-DNA kinase activity and a 3’-DNA phosphatase activity. It has three domains, a C-terminal kinase domain, a central phosphatase domain, and an N-terminal forkhead associated (FHA) domain that is responsible for protein-protein interactions. Reduction in expression of PNK by RNAi sensitizes cells to ionizing radiation and topoisomerase I inhibitors.
Purification: Purified against PNK protein affinity chromatography.
Recommended Dilutions: Western Blotting 1:2000; ELISA 1:1000
Storage buffer: Tris Buffer pH 7.4, containing 50% Glycerol without azide.
Storage instructions: Store at -20°C (Recommended).
*N. B. (Membrane blocking): One hr at RT in 5% (w/v) non-fat milk; (Primary Ab): Overnight at 4°C in 1% non-fat milk; (Secondary Ab): One hr at RT in 1% non-fat milk. All incubations and washings are in 1X TBS, 0.1% Tween-20.
